Eugene Gladstone O'Neill, 16 Ekim 1888 tarihinde New York, ABD'de dünyaya gelmiştir. Döneminin ünlü tiyatro oyuncusu James O'Neill ile Mary Ellen Quinlan'ın oğlu olan yazar, çocukluğunun büyük bir kısmını babasının turneleri nedeniyle otellerde ve trenlerde geçirdi. Princeton Üniversitesi'ne girmesine rağmen kısa süre sonra eğitimini yarıda bıraktı. Gençlik yıllarında denizcilik yaptı, çeşitli işlerde çalıştı ve bu dönemde ağır depresyonla mücadele etti.
